Lipid imaging mass spectrometry (LIMS)

Imaging mass spectrometry is a new and revolutionary technique that enables, for the first time to visualize the distribution of molecules (proteins, metabolites, drugs… etc.) in tissue sections. It presents several variants but in its MALDI (matrix assisted laser desorption/ionization) version, the process starts with a fresh-frozen biological sample that is sectioned with the aid of a cryomicrotome. After being covered with a suitable organic substance (the “matrix”), it is introduced in the ionization chamber of a mass spectrometer, where it is scanned at a pre-defined spatial resolution. Integration of the m/z in the mass spectrum enables reconstructing the spatial distribution of all the molecular species detected. The technique presents a great robustness, sensibility and reproducibility. In each experiment, it is possible to determine the spatial distribution of hundreds or even thousands of molecular species without any previous labelling.

Due to the strict regulation of the lipid profile of the cells and its alteration in the context of a pathology, this technology enables identification of novel lipid biomarkers for screening of pathological samples. In the GS2M we have developed new protocols, instrumentation and software, making a measurable impact in the advance of this technology. Our research involves collaborations of national and international hospitals, research groups and companies.
